Setting up VCS root for TFS 2010
I have setup a TeamCity and TFS 2010 Sp1 server. Both hosted on separate machines and I have installed Team Explorer 2010 on Teamcity server. In my TC project setup when I hit "Test Connection" on Adding a NEW VCS root, test fails with below error.

ERROR - No TFS assemblies were found on the system. Please make sure you have Microsoft Team Explorer 8.0, 9.0, 10.0 or 11.0 installed

Have you restarted TeamCity server after Team Explorer installation?
Error messages in teamcity-server.log and teamcity-vcs.log files can provide more details.
